Monitoring endoplasmic reticulum quality control by Rashmi Tripathi
Quality control of proteins being expressed in the endoplasmic reticulum (ER) is critical for the normal functioning of a cell and disruption of this process is observed in various diseases like Alzheimer's disease, Retinitis pigmentosa (RP), diabetes etc. Monitoring quality control in the ER might enable the development of effective therapeutic strategies for treatment of such diseases. Confocal imaging of cells in the Department of Chemical Engineering and Biotechnology has enabled the study of this phenomenon in greater detail. The first image shows co-expression of GFP (green) and mCherry (red) in the ER (yellow when GFP and mCherry are colocalised) in a normal cell. In the the second image expression of GFP-tagged polyglutamine protein (green) that is prone to aggregation and mCherry (red) is also targeted to the ER. The ER network is no longer visible and is replaced by large aggregates throughout the cell, including the nucleus (stained blue), indicating cell stress.
